My target calorie deficit is 400. Do I need to try and keep it there throughout the day, or just have a 400 deficit at the end of the day?

What matters is your deficit at the end of the day-some people may find it easier to control their numbers by managing them over the course of the entire day. When pursuing muscle gain, you need to eat frequently throughout the day, and all meals should contain protein, carbs and fat, which should keep you in a small deficit regularly.
